Adenovirus ELISA kit description:

Diagnostic Automation Adenovirus ELISA test kit is an in vitro procedure for the qualitative determination of adenovirus antigen in feces. It is a double antibody (sandwich) ELISA using a polyclonal anti-adenovirus antibody to capture the antigen from the stool supernatant.

Material Provided with Adenovirus ELISA Kit:
1. Test Strips Microwells: 96 test wells containing anti-adenovirus polyclonal antibodies
2. Reagent 1: anti-adenovirus monoclonal antibodies
3. Reagent 2: anti-mouse antibodies HRP-conjugated
4. Positive Control: diluted adenovirus antigen
5. Negative Control: buffer with Thimerosal
6. Chromogen:TMB and peroxide
7. Wash Concentrate, 20x
8. Stop Solution: 5 % phosphoric acid

Materials Required, Not Provided:
1. Precision pipettes
2. Distilled or deionized water
3. EIA kit Microplate Washer
4. EIA kit Microplate Reader with a 450 nm/620 - 650 nm filter

Adenovirus Elisa Kit Background Information:
Acute diarrheal disease in young children is a major cause of morbidity worldwide and is a leading cause of mortality in developing countries. Research has shown that enteric adenoviruses, primarily Ad40 and Ad41, are a leading cause of diarrhea in many of these children, second only to the rotaviruses. Infections are most frequently seen in children under two years of age, but have been found in patients of all ages. Further studies indicate that adenoviruses are associated with 4 - 15% of all hospitalized cases of viral gastroenteritis. Adenoviruses have an incubation period of 8 - 10 days, followed by viral shedding for an approximate period of 7 - 14 days. The main symptoms are diarrhea and vomiting, however a fever is also seen in 40 - 90% of the cases. The diarrhea resulting from enteric adenoviruses is longer in duration than that caused by the rotaviruses, usually lasting 7 - 8 days. This is a leading reason why patients seek medical attention for this condition. Viral transmission is believed to be by the fecal-oral route.

Adenovirus (Fecal) ELISA Test Principle:
During the first incubation, adenovirus antigens present in the stool supernatant are captured by
antibodies attached to the wells. The second incubation adds an additional anti-adenovirus antibody that sandwiches the antigen. The third incubation attaches horseradish peroxidase to the sandwich. After washings to remove unbound enzyme, a chromogen is added which develops a blue color in the presence of the enzyme complex and peroxide. The stop solution ends the reaction and turns the blue color to yellow.

Notable features of the Parasitology ELISA Kits:
User-friendly directions and explanation of test procedures
Simple and safe reagent preparation
Clear instructions on specimen collection
Comprehensive package of required materials
Explicit quality control and storage guidelines
Reliable and easy-to-read test results

Product inserts for most Parasitology ELISA kits follow a similar method. See the Adenovirus ELISA Kit product insert for details on preparation, procedures, quality control, and test result interpretation.
